Studies on the Anther Culture of Garlic (Allium sativum L.)


Abstract
Anthers of garlic cv. Nagano White, Jaeju, Sanghai Early were cultured on MS, LS and B5 media supplemented with several growth regulators and glutamic acid under a 16hr regime at 25¡É. 1. Callus was observed about 25 days after inoculation, but embryoid was not found.
2. The optimal pretreatment for the callus induction was 5¡É/12hrs.
3. Callus induction was strongly dependent on the presence of NAA while IAA or 2, 4-D was not effective.
4. Bs medium was higher than LS medium in the ratio of shoot differentiation, especially shoot differentiation was more effective at higher concentration of kinetin in BS medium and low concentration of BA in LS medium.
5. Adventitious shoots developed roots in 2 wks on rooting medium(1/2MS) and produced about 5 bulblets per callus 4 wks after inoculation. Then, shoots emerged from the bulblets 9 wks after inoculation.
6. Varietal difference in the anther culture was clearly evident. `Nagano White¢¥ was well responsed to the inducible treatment used in this experiment, and `Jaeju¢¥ was intermediate, but `Sanghai Early¢¥ not responsed at all.
7. The chromosome observation revealed that the callus was mixoploid including both haploid and diploid, but the root tips of regenerated plantlets proved to be diploid.
